        AimGel Mouse T Cell Activator presents anti-CD3 and anti-CD28 signals on soft hydrogel microparticles built to the size of a real mouse T cell (7-12 µm). The fluid lipid membrane lets those signals move freely across the surface, the way they do on a natural antigen-presenting cell. Activation is gentle and effective, whether you start from isolated mouse T cells or straight from PBMC.

        Specifications

        A suspension of AimGel hydrogel microparticles made of chemically crosslinked dextran. Microparticles sized ~7-12 µm, coated with a phospholipid bilayer, with mouse T cell activating signal panels docked to the membrane.

        Viability, purity and activation

        Mouse T cells activated with AimGel, starting from mouse PBMC. Left: cell viability remained between 80% and 92% across days 3, 5 and 7. Centre: CD3+ fraction of live cells rose from ~14% at day 0 to ~98% by day 5, showing selective expansion of T cells without a separate isolation step.         What about second stimulation?

        Thanks to gentle and biomimetic signal presentation, AimGel for mouse T cells is able to effectively restimulate the expansion of mouse T cells. Mouse PBMC were initially stimulated with AimGel for mouse T cells, and were then restimulated with AimGel for mouse T cells on day 11, except for the negative control.

      • FAQs

        Can I start from mouse PBMC, or do I need isolated T cells?

        Either. AimGel selectively expands CD3+ T cells: starting from mouse PBMC at ~14% CD3+, the population reaches ~98% CD3+ by day 5, so a separate isolation step is often unnecessary. See the protocol for seeding guidance for each starting material.

        How do I remove the AimGel microparticles?

        Add the included lysis buffer. Dextranase fully degrades the microparticles in 30 minutes at 37 °C, without affecting your cells. See the protocol for timings.
        If you need AimGel that clears on its own at a set point, we can formulate it to hydrolyse under your culture conditions, get in touch.

        Can I re-stimulate my mouse T cells?

        Yes. Growth typically slows after day 10, and re-stimulating promotes continued expansion with minimal activation-induced exhaustion. We recommend an AimGel-to-cell ratio of 0.25:1 for re-stimulation.
        Do not use the lysis buffer if you plan to re-stimulate, as residual dextranase will degrade freshly added AimGel unless it is thoroughly washed out.

        Do I need feeder cells?

        No. AimGel is a fully synthetic, feeder-free alternative with no animal components.
